About the Project

This is an exciting and unique opportunity for an ambitious student researcher to join the laboratory of Dr. Oran Kennedy, on a 4-year PhD studentship, in the emerging area of joint injury/disease, mechanobiology, inflammatory processes in post-traumatic osteoarthritis (PTOA) and novel drug delivery. The Kennedy laboratory is located within the Tissue Engineering Research Group (TERG) cluster at the 123 St. Stephens Green campus. Dr. Kennedy is also a PI in the Trinity Center for Bioengineering (TCBE), and has a variety of ongoing projects with researchers there, as well as in the Advanced Materials and BioEngineering Research (AMBER) Centre which brings together leading research centres in bioengineering and nanotechnology.

Overview of project:

Arthritis is a condition that is typically associated with advancing age however a subset of the disease called Post-Traumatic Osteoarthritis (PTOA) affects a much younger population. PTOA develops following joint injury, such as ACL rupture of the knee, about 10 years after injury but why this happens is unknown. What has become clear in recent years is that other joint tissues (in addition to cartilage) are involved, and that inflammatory processes are also involved in addition early intervention is crucial. The aim of this project is to use novel models (in vitro and in vivo) to understand the role of different joint tissues in PTOA initiation and progression and to develop ways to target those specific tissues with drug delivery systems that can mitigate joint degeneration.

The student will be primarily based at RCSI and will work closely with members of a multidisciplinary project team including PIs, postdoctoral and postgraduate researchers within this research cluster. Opportunities will also arise for working with national and international collaborators (particularly in the New York region).

The purpose of the PhD is to determine the means by which bone and cartilage tissues communicate, or cross-talk, following joint injury in such a way as to initiate joint disease. Furthermore, we wish to understand how treatment paradigms can be improved in order to target specific tissues and thus prevent disease. The project will utilise state-of-the-art techniques such, in vivo mechanical loading models, mechanical/materials testing, advanced skeletal imaging (including micro-computed tomography and in vivo multi-photon microscopy), advanced histological techniques, RNA sequencing, bioreactor & explant culture systems, fluorescence and electron microscopy. Experience working with pre-clinical GMO models would also be an advantage.

Mandatory Specification:

Upper Second Class (2.1) honours degree (or equivalent) in a relevant subject.

Desirable Requirements:

Excellent technical writing skills

Materials/mechanical testing

Advanced skeletal imaging (including micro-computed tomography and in vivo multi-photon microscopy)

Histological techniques,

Cell/molecular biology

Fluorescence/electron microscopy.

Experience working with pre-clinical GMO models would also be an advantage.
