今天介绍的是山农杨克强-房鸿成团队发表在The Plant Journal的研究论文《JrWRKY21 interacts with JrPTI5L to activate expression of JrPR5L for resistance to Colletotrichum gloeosporioides in walnut》。
核桃炭疽病是由炭疽菌引起的危害我国核桃产业的重大病害。虽然WRKY转录因子在植物免疫中起着关键作用,但WRKY在核桃抗炭疽病中的作用尚不清楚。
在本研究中,作者通过转录组测序和qRT-PCR鉴定了一个差异表达基因JrWRKY21,该基因在核桃感染球孢霉后显著上调。
如病毒诱导的基因沉默和瞬时基因过表达所示,JRWKY21正调节核桃对球孢霉的抗性。此外,JrWRKY21在体外和体内直接与PR基因JrPTI5L的转录激活子相互作用,并可与JrPTI5L启动子中的W-box结合进行转录激活。JRPT5L可以通过结合启动子中的GCCGAC基序诱导PR基因JrPR5L的表达。
本研究数据支持JrWRKY21可以通过WRKY21-PTI5L蛋白复合物间接激活JrPR5L基因的表达,以促进核桃对球孢霉的抗性。研究结果将有助于加深对核桃抗病机理的认识,并通过分子设计育种培育抗炭疽病品种,促进核桃的遗传改良。
Walnut (Juglans regia L.) anthracnose induced by Colletotrichum gloeosporioides is the catastrophic disease impacting walnut industry in China. Although WRKY transcription factors play a key role in plant immunity, the function of WRKY gene in walnut resistance to C. gloeosporioides is not clear. Here, we identified a differentially expressed gene, JrWRKY21, through transcriptome sequencing and qRT-PCR that was significantly upregulated upon C. gloeosporioides infection in walnut. JrWRKY21 positively regulated the walnut resistance to C. gloeosporioides as demonstrated in virus-induced gene silencing and transient gene overexpression. Additionally, JrWRKY21 directly interacted with the transcriptional activator of PR gene JrPTI5L in vitro and in vivo and could bind to the W-box in the JrPTI5L promoter for transcriptional activation. And JrPTI5L could induce the expression of PR gene JrPR5L through binding GCCGAC motif in promoter. Our data support that JrWRKY21 can indirectly activate the expression of JrPR5L gene by WRKY21-PTI5L protein complex to promote resistance against C.gloeosporioides in walnut. The results will enhance our understanding of the walnut disease resistance mechanism and facilitate the genetic improvement of walnut by the molecular design breeding for anthracnose resistance varieties.
