“A Zika virus-specific IgM elicited in pregnancy exhibits ultrapotent neutralization”
先天性寨卡病毒(ZIKV)感染导致高达14%的感染ZIKV母亲所生婴儿的神经发育缺陷。中和抗体是保护性免疫的关键组成部分。来自美国国立卫生研究院的Mattia Bonsignori以及康奈尔医学院的Sallie R. Permar团队合作证明血浆IgM有助于妊娠期ZIKV免疫,在出现症状后3个月内介导中和反应。从一名感染ZIKV的孕妇中,研究者分离出一种五聚体ZIKV的特异性IgM(DH1017.IgM),其表现出依赖于IgM同型的超强ZIKV中和作用。DH1017.IgM靶向病毒包膜的结构域II形成的二聚体表位。病毒上的表位排列与DH1017.IgM的所有十个抗原结合位点的同时结合相兼容。相比于之前只有IgG表达时,DH1017.IgM在致命ZIKV攻击时更有效的保护小鼠对抗病毒血症。本研究确认了IgM同型抗体在预防寨卡病毒感染方面的作用,并确定了DH1017.IgM作为一种安全有效的候选免疫疗法,特别是在怀孕期间。
