JIA | 中国农科院北京畜牧兽医研究所牟玉莲课题组鉴定一个新的猪基因组友好基因座

外源基因在转基因动物和细胞中稳定、高效的表达,对于基因功能的研究、生物反应器和医学模型的建立至关重要。动物基因组中的友好基因座能使外源基因高效稳定的表达且无副作用,但目前猪基因组中可供外源基因安全、高效定点整合的基因座相对较少,限制了多转基因猪的研究和发展。
近期,中国农业科学院北京畜牧兽医研究所牟玉莲课题组提出一种将猪I型胶原α1链(collagen type I alpha 1 chain,COL1A1)基因座作为友好基因座表达外源基因的新策略。该研究为外源基因在猪基因组中稳定表达提供了新位点,可广泛应用于基因功能研究、多基因聚合生物育种材料研发、生物反应器和医学模型建立等多个领域。相关研究成果在Journal of Integrative Agriculture (《农业科学学报》(英文),JIA) 在线发表。
该研究通过荧光定量、蛋白质印记、转录组测序以及细胞增殖实验等多种技术手段,在多个水平上证明将外源基因定点整合在COL1A1基因座不影响COL1A1基因本身的转录和翻译,不改变内源性基因的整体表达,同时对细胞增殖无显著影响。该研究表明COL1A1基因可作为猪基因组中外源基因定点整合的友好基因座。
